An antigen test is a blood test used to detect the presence of antibodies to a specific antigen. The test is done by adding a serum containing known amounts of antigens to a sample of blood. If antibodies are present in the serum, they will bind to the antigens in the sample. The amount of antibody bound can be measured and used to determine whether or not an individual has been exposed to the antigen.

The antigen test is often used to screen for exposure to certain viruses, such as HIV and hepatitis B and C. It can also be used to diagnose autoimmune diseases, such as lupus and rheumatoid arthritis. In addition, the antigen test can be used to determine if someone has received a vaccine, such as the MMR vaccine.

One of the earliest antigen tests was for syphilis, a sexually transmitted disease. In 1905 August von Wasserman developed a test using blood serum from an infected individual which could diagnose early syphilis infections in blood donors before the appearance of symptoms. The test was used to screen individuals who were donating blood in much the same way that modern tests are used today.

The first application of this type of testing in forensic science was in 1915 when it was used to help discover whether or not poisoning had taken place in cases where no poison could be found. Today, antigen tests are commonly done on samples at crime scenes when looking for evidence of biological fluids such as semen, saliva and other bodily fluids. This is especially useful in rape cases when there is little other evidence to work with.

Most antigen tests are done using ELISA, the enzyme-linked immunosorbent assay. Antigens and antibodies can be tagged with enzymes which make them easier to detect and measure. Many different types of antigens can be tested including blood group antigens, tumor markers and drugs such as cocaine and certain antibiotics. If an antigen test suggests that a person has been exposed to a particular organism then further testing such as nucleic acid amplification or PCR may be used to identify the specific microbe.

Antigen testing will regularly produce false positives for individuals who have recently received vaccines containing the relevant antigens, giving rise to fears about vaccinations. Normal blood cells may also give a false positive result. In order to avoid these problems, the antigen test is often used in combination with other tests, such as the antibody test.

The antigen test is a valuable tool in modern medical diagnostics. It can be used to screen for a variety of diseases and disorders and can provide important information in forensic investigations. The test is reliable and relatively easy to perform, making it an important part of modern medicine.
